When using JScan adjusting the tire size, does it change the shift points in the transmission at the same time or is there another place I need to go?

No software, no tazer, does that. The truck's systems adjust things as needed, if needed.
All you need to do is change tire size. There's nothing else anything outside of the truck does to shift points.
Shift points are based on so many things, it's not anything an amateur should attempt to deal with anyway. That's for tuners.

Just put in the correct tire size and verify with speedometer/odometer and GPS.
