What you are describing- I would drain the diff fluid in the rear and check the backlash on the ring gear. (Just my .02)

The only other thing coming to mind would be bias ply or directional tires that are mounted backwards would be why you are hearing noise when coming off the throttle.
Checking backlash isn’t overly complicated. Open the pumpkin, stick a dial indicator on and push the gear set back and forth to measure slop. ?

Does not need to be on a bench….
